Was the Spit 5b Merlin fitted with the Hooker two stage supercharger?

The Hooker redesigned supercharger was introduced, as you said, on the two-speed Merlin XX, but also on the single-speed Merlin 45, so the answer is partially yes, the Vb did have the Hooker supercharger.
Two-stage was a later development altogether.

Red’s question was about two-stage supercharging.
That was introduced on the 60 series – Spit Mk VII, VIII and IX (but not in that order ;))

Quick check shows it was the Merlin 60 series onwards that carried the two-speed, two-stage supercharger

No. The two-stage supercharger was the key improvement over the Vb enjoyed by the Spitfire Mks IX and VIII
